Consider the following mutations in a C elegans gene whose gene product is a protein required for DNA replication. Indicate the consequence of the mutation to transcription of the gene and phenotype of C.elegans.
| Transcription | Phenotype of C. elegans (assume Homozygous for mutation) | |
| Mutation of TATA Sequence: | ||
| Mutation in Splicesome binding site | ||
| Mutation within protein coding region | ||
| A 4- base-pair deletion on the 5\' UTR region: |
Solution
Mutation of TATA sequence: Transcription is halted due to inability in the binding of transcription factors, the phenotype would be a loss of production of protein.
Mutation in spliceosome binding site: Splicing of nascent mRNA into mature mRNA is halted, the phenotype would be no production of mature mRNA.
Mutation within protein coding region: Transcription process is not affected, phenotype would be either truncated protein or altered protein.
A 4 base pair deletion on 5\' UTR: Transcription process is not affected, phenotype would be absence of the protein or altered protein production (5\' UTR is important for the regulation of translation of a transcript)
