No, it not at all mean that these two amino acids cannot be present in the protein sequence. Other than the sequence of the shine dalgarno it also depends upon the position on where it placed on the gene ie 5\' end as a promoter from where the enzymes can bind process can start..!! So along with the sequence promoter other factors also varies...
