Record the brand of lactase, number of units of lactase/tablet and the expiration date. Weigh one lactase tablet. Note whether the tablet Is 9000 FCC (Food Chemical Codex) or 3000 FCC units/tablet. Grind a tablet using a mortar and pestle until finely ground. Suspend/dissolve to 1000 units/mL in 0.01 M PO_4 buffer, pH 7.0. Dilute 1:1000 with 0.01 M PO_4 (pH 7.0). The total volume of diluted enzyme should be 10 mL. You will use this diluted enzyme In both parts 2 and 3. Keep on ice.

Solution

Generally, tablets contain 9000 FCC units for their medication use. To convert 9000 units into a solution at 1000units per ml, we use following formula

U1 x V1 = U2 x V2 where U is units and V is the volume.

Here we take 9000 units of tablet is dissolved in 1 ml of water.

=9000 x 1 = 1000 x V2

V2 =9 ml

Therefore, we need 9 ml of water to make 9000 units of tablet into 1000units/ml solution.

We need to make dilution in such a way that it takes final 10 ml solution after diluting in 1:1000 dilutions. For that, we use formula here:

P2 x V1 = P1 x V2, Where, for the initial volume, we take 1 proportion and 1000 is taken as 2nd proportion.

= 1000 x V1 = 1 x 10 ml

V1 = 10/1000 = 0.01ml

Therefore, we need 0.01ml of sample volume and 10-0.01 = 9.99 ml of diluents to make the solution of 10ml into 1:1000 dilutions.
