Step1) Prepare agar nutrient medium and autoclave it.

Step 2) prepare arabinose and ampicillin by adding 3 ml of transofrmation solution to both the substance.

Step 3) label 3 petri plates a) LB media b) LB+AMPLICILLIN c) LB+AMPICILLIN+ARABINOSE

Step 4)Pour the nutrient agar according to the label in the petriplates

Step 5) Hydrate lyophilized Ecoli HB101 by adding tranformation solution

Step 6) Streak the plates with bateria

Step 7) make the pGLO plasmid.

Step 8) add 1 ml of transfection solution {buffered saline solution (HeBS) containing phosphate ions is combined with a calcium chloride solution containing} to LB broth

Step 9) culture the petriplate with positve and negative pglo control.

Step 10) scan under UV light.
