Several mutants of Neurospora arc isolated, all of which require compound G for growth. The compounds (A to E) in the biosynthetic pathway to G arc known, but their order in the pathway is not known. Each compound is tested for its ability to support the growth of each mutant (I to 5). In the following table a plus sign indicates growth and a minus sign indicates no growth. A. What is the order of compounds in the pathway, and at what point in the pathway is each mutant blocked? B. Would a heterokaryon composed of double mutants 1.3 and 2.4 grow on minimal medium? C. What about a heterokaryon of the double mutants 1.3 and 3, 4? D. What about a heterokaryon produced by mixing double mutants 1.2 and 2.4 and 1.4? PCR was used to amplify segments of DNA from two different genes (A and B) using genomic DNA from two different individuals (P1 and P2) as templates. Four bands were produced from each individual, indicating that they were heterozygous for long and short alleles of each gene.

Solution

A)Sequence of the pathway is as follows; C B A D E G

Blockage in murant 1 is at A to D conversion

Mutant 2: Blockage at E to G conversion

Mutant 3: blockage at D to E conversion

Mutant 4: blockage at C to B conversion

Mutant 5:block at B to A conversion

B)

Several mutants of Neurospora arc isolated, all of which require compound G for growth. The compounds (A to E) in the biosynthetic pathway to G arc known, but
