a. The plasmid concentration is 0.002 ug/ul or 2 ng/ul. If 10uL of plasmid is taken in both the tubes which makes it 20 ng of plasmid DNA in both +pGLO and -pGLO tubes.
b. The ara in the first plate is acting like a selection marker which makes the colonies which have this gene are able to survive.
