This article speaks almost exclusively about the hypoxia sense and response mechanism based on HIF-1 and related proteins. The article does not go into detail about the laboratory techniques used in detail except the use of transgenic animals and mutants. Semenza and Ratcliffe individually used transgenic mice to study the role of HIF in regulation of erythropoetin gene expression.

Kaelin had used VHL mutants to understand vascularization in renal cells (note that it is specified he observed vascularization in cancer cells with VHL mutation this could be either intentionally induced by the researcher or can also be a sharp observation).

Although not specified in the article I would say they researchers would have extensively used immunohistochemical techniques to understand the molecular interactions between the proteins and between the protein and DNA. And also it is possible that they studied protein and gene expression with several PCR techniques to understand the influence of different factors on the level of expression of important proteins such as VEGF and erythropoetin.

And offcourse they employed several animal cell culture techniques in various media conditions to understand the effects on metabolism.

Personal note : I greatly appreciate and admire the elegance of this molecular process which I was not aware of, the researchers are worthy of every award bestowed.
