A linear DNA molecule is subjected to complete restriction digestion by (1) EcoR1 restriction enzyme alone, (2) Bam HI restriction enzyme alone, and (3) both enzymes together. The fragments are run on an agarose gel. Results of the stained gel are shown below (the size standards are shown on the right side of the gel).

Solution

Choose any one of the columns and add all the fragment lengths

EcoR1 = 10+5+2 = 17Kb

check with the other columns also

BamH1 = 9+8 = 17Kb

Both = 9+5+2+1 = 17Kb

There are no two fragments of the same size by both enzymes.
