Assume you count 250 colonies on the +pglo LB/amp/ara plate and a confluent lawn of bacteria in the +pGLO LB/amp plate. Did you expect this result? If not, give a detailed explanation! How could you prove your theory?

Solution

The + sign indicates that the plasmid has got inserted in the bacterial cell. The transformed bacterial cell will be able to survive in the presence of ampicillin.

However, the given results are not expected as there wont be difference in growth pattern provided the transformed culture plated is same.

Arabinose is a sugar which activates the gene for GFP production (Green Fluorescent Protein). On LB/amp plate it will show growth but no fluorescence due to absence of ara whereas on LB/amp/ara there will be growth as well as fluorescence. the only difference will be in glowing of colonies. Both the plates should show same extent of growth.
