Previous studies have shown that activation of NMDA receptor
Previous studies have shown that activation of NMDA receptors, depending on their location in the brain, can either activate or repress transcription of genes that contain cAMP responsive elements (CRE) in their promoters. Mouse fibroblasts, which do not express N-cadherin, were transfected with a control plasmid with no insert (vector) or with a plasmid containing the domain of the N-cadherin gene encoding its cytoplasmic terminal fragment (N-Cad/CTF2). The effect of overexpressing this domain on transcription of two genes, c-fos and graph, was then determined by RT-PCR and analyzed after 29, 35 and 38 cycles of amplification. What can you conclude about a signaling cascade, c-fos, graph, and the Terminal fragment of N-cadherin from these studies? If you were to examine c-fos and graph expression in neurons of PS1 -/- knockout mice, how would you predict expression patterns to compare to those of wild-type mice?
Solution
C. overexpression of N-Cad(CTF2) has the negative effect on the expression of C-fos. It might possible that N-Cad(CTF2) overexpression is regulating the c-fos expression as a result of it C-fos expression level is decreased. whereas overexpression regulates the GAPDH gene positively is used as a reaction control as we can see from the RT-PCR band.
