Assume the density of plasmid-containing cells you had when you began the exercise was 108 cells/mL. How many copies of E. coli chromosomes were in the microcentrifuge after lysing the cells? Remember your tube contained 1mL of cells which had been pelleted.
Solution
There is one single circular chromosome present in each cell of E.coli unlike that of human which has 23 pairs of it. Now as the tube sample contained 1 ml of cells which had 108 E.coli cells in it, so after microcentrifuge 108 copies of chromosome will be found.
