Suppose you are investigating an uncharacterized fungal spec

Suppose you are investigating an uncharacterized fungal species and identify a novel amino acid sequence that is highly conserved among every nuclear protein you have examined in this organism. You suspect this sequence is a nuclear localization sequence How could you show that your hypothetical NLS is both necessary and sufficient for import of proteins into the nucleus in this species?

Solution

the corresponding codon region in the mRNA may be manipulated or mutated to change the codon sequence. this results in a different amino acid sequence in the proteins. the transport of the proteins into the nucleus can be monitored.

the amino acid sequence in the polypeptide chain can be modified by deletion of some amino acids.

the corresponding region in the DNA can be induced by a framshift mutation or single nucleotide deletion,, so that the entire region of amino acids woud change.
