The purpose of ERK protein in a western blot is to use it as a positive control. The kit for Western blot is validated for the detection of ERK 1/2 protein phosphorylated at threonine 185 and tyrosine 187 and ERK 1/2 total. Hence for this both the anti-Erk solution and anti-phospho Erk antibody is used.
