You are working in the lab late one night and you accidentaly mix a few protein samples together. You know the identity of the proteins that you might have mixed and you also have antibodies available that are specific for each protein.Explain how you can confirm which proteins are present in the mixed sample.Provide brief details of the experimental technique used.
Solution
If we know the names of antibodies itself is enough to tell the names of proteins in the mixture of proteins. Because antibody react with specific epitope of a particular protein. Hence it is easy to say which protein present in samples.
The techniques used to identify this are dot blot, western blot, ELISA are some of them.
Dot blot:- is a simple method where proteins spotted on a membrane and apply the antibody, if concern protein present in the dot of sample it react with it and using secondary antibody we can find the signal.
Western blot:- similar to dot blot but here proteins are denatured and loaded into a SDS-PAGE then transferred onto a mrmbrane the rest of the things are similar to dot blot.
ELISA:- here the proteins directed by antibody in a perticular concentration. Whole principle is similar to dot blot and western blot.
