What is the expression system used to generate a fusion protein? what is a fusion protein and its purpose?
Solution
To generate a fusion protein various expression systems can be used, such as:
1. Bacterial system
2. Yeast system
3. Baculovirus system
4. Mammalian vector system
Different type of plasmid containing different promoters are used in each of these systems. For bacterial system T7 promoter system is used in common.
Fusion protein is a recombinant protein generated when extra amino acids are attached in C or N terminal of the protein. These amino acids ac as various tags such as His tag, Strep tag, EGFP tags etc. The pupose of these tags are given below:
1. Helps in solubilization of the protein.
2. Helps in purification of the protein by affinity chramatography
3. Helps in localization studies
4. Helps in detection of the protein present by using westerns blotting.
5, Acts as reporter gene for identification of the protein present
