Difficulties in purification of taq from its original source T.aquaticus :-
Availibility of Thermous aquaticus: The temperature of an environment can be considered as one of the most influencial factors for T. aquaticus. Because it can survive only at temperatures of 55-100 degrees celsius in weakly acidic to alkaline (pH 5-9) waters. As these organisms exist at high temperatures it is found in hot water spring, marine thermal springs, low saline solfataric springs and thermally polluted waters. If this temperature and pH do not found in a perfect range it will never grow.
Maintaining temperature – At the time of purification of Taq from T. aquaticus in lab, the temperature of T. aquaticus should be maintained properly. Because at low temperature it will not alive.
Salinity: T. aquaticus is very sensitive to changes in salinity. Its growth is inhibited even if there is a 0.5% concentration of NaCl and cannot grow in any system with NaCl concentrations above 1%. So perfect salinity must be required for their growth
In a nutshell we can say that the above conditions are required to grow T. aquaticus. If these are not available properly T. aquaticus will never sustain in envioronment and as a result purification of taq become so difficult.
