What is the difference between sizeexclusion chromatography
What is the difference between size-exclusion chromatography and ion exchange chromatography?
Solution
A. Size excl usion chromatography – It is als known as (gel filtration) and it divides proteins basic on molecular size . Can differentiate between higher- ordered and monomeric structures. If the protein (e.g. 80kDa) is bit of a huge compound, it will fractionate at a size prominent than 80kDa (on a denaturing SDS-gel it will fractionate at 80kDa).
B. Ion exchange chromatography – Divides proteins basic on charge . The overall accusation of a protein is the sum of all of the accusation on the amino acids at a specific pH. When these accusation are equal this is known as the isoelectric tip. At pH > isoelectric tip proteins have a net negative accusation, at pH < isoelectric point they have a net positive accusation.
