PLEASE EXPLAIN THE ANWER
21. You calibrate a size-exclusion column which was made using Sephadex G-100. Sephadex G-100 is a size exclusion resin with a fractionation range for proteins of 4,000-150,000 daltons. The elution profile (A280 vs. fraction number) is shown below and the standard proteins you used to calibrate the Sephadex G-100 column are listed in the table below. 280 Fraction Number MW (daltons)standards: Peak C represents which of your protein Proteirn Catalase Chymotrypsinogen Cvtochrome c Myoglobin Myosin Serum Albumin 222,000 23,240 13,370 16,900 524,000 68,500 A. Basics
In size exclsion chromatography the protein with high moleclar weight eltes first and smaller elutes later.
You have used six standard protein but chromatogram shows only five peak. The order of elution for yor standard protein is
1. Myosin
2. Catalase
3.Serum albumin.
4. Chymotrypsinogen
5. Myoglobin.
The above order is based on moleclar weight of protein. So the peak C belongs to serum albmin.
