1)200mM Sodium Hydroxide /1% SDS
Solution
1) 200mM Sodium hydroxide : It is used in alkaline lysis step of plasmid isolation.Sodium hydroxide helps to denature the genomic and plasmid DNA.It also denatures the cellular proteins that maintain the cell membrane structure.
2)1% SDS:It is also used in alkaline lysis step of plasmid isolation. SDS detergent causes cell lysis by solubilising the phospholipids and proteins of the cell membrane thereby releasing the contents of the cells.
