Determine the primary sequence of the polypeptide, many individual chains it is made up of, and if/where disulfide bonds may exist.

Amino acid analysis revealed the following composition for the decapeptide - Arg, (Cys)2, Gln, Gly, Lys, Met, Pro, Ser, Tyr

1. Treatment of the heptapeptide with chymotrypsin resulted in no cleavage. Treatment of the heptapeptide with pepsin resulted in a dipeptide and pentapeptide.

2. Treatment of the heptapeptide with trypsin resulted in the generation of free arginine, a dipeptide, and a tetrapeptide.

Solution

Proteins are the most abundant organic molecules in animals, playing important roles in all aspects of cell structure and function. Proteins are biopolymers of acids, so named because the amino group is bonded to the carbon atom, next to the carbonyl group. The physical and chemical properties of a protein are determined by its constituent amino acids. The individual amino acid subunits are joined by amide linkages called peptide bonds. Figure shows the general structure of an acid and a protein.
