Solution

Answer:

Hybridization: It is the act of mating different species of organisms to create a hybrid (New modified variety of organism). Here, a double stranded nucleic acid is formed by combining two complementary strands of DNA/RNA.

Stringency: It is the limit or extent to which hybridization can occur between nucleic acid with mismatched sequences.

** Two ways to decrease the stringency of hybridization is by:

1.  Increasing NaCl concentration.

2. Reducing temperature below the temperature of molecules.

The stringency level is determined by the hybridization temperature and the salt concentration in the hybridization (high temperature and low salt is more stringent as only perfectly matched hybrids will be stable). In case of probes that don\'t match the target completely, the stringency must be reduced to a level that allows imperfect hybrids to form. If the stringency of the hybridization is too low, then the probe may bind to too many sequences to be useful.
