Solution

Aminoacids are classified based on their properties,side chain and size.

Hydrophobic aminoacids: Glycine, alanine, valine, leucine, isoleucine, proline, phenylalanine, tryptophane and methionine

Positively charged aminoacids :  lysine, arginine, histidine

Negatively charged aminoacids : aspartate, glutamate

Polar aminoacids : serine, threonine, asparagine, glutamine, cysteine and tyrosine

Amino acid substitution may be significant if there is no change in the protein structure and function. If a polar aminoacid is replaced by another polar aminoacid or by the aminoacid with same property and size.

But the amino acid substitution may not be significant if there is a change in the protein structure and function. If a positively charged amino acid is replaced by negatively charged amino acid, it affects the protein function. If a larger amino acid is replaced by smaller aminoacid, it affects the protein structure.
