ANS-Methylene blue used for the bacterial identification and morphology and when this stain contact with the acids it gives deep blue color, methylene blue stain the Nucleic acid and bind with the DNA .its a basic dye and can easily bind with the DNA.methylene blue is also used as a indicater if the cell are dead or alive.alive cell do not stain but dead cell easily stain with the methylene blue.

Idodine react with the starch and it gives the purple color which indicate the concentration of starch and glycogen into the cell and facilitate the staining of nucleous.
