Km is the Michaelis-Menten constant that shows concentration of the substrate when the reaction velocity is equal to one half of the maximal velocity for the reaction. Km measure how well a substrate complexes with a given enzyme,also known as its binding affinity.

                                 Km = [S],      r = 1/2 Vmax

Km is equal to that concentration of substrate at which rate of formation of product is half the maximum rate obtained at high concentarion of substrate.
