Why would a vector/insert ratio of 3:1 generate the most recombinants. And yes the ratio is vector to insert 3:1 not the other way around

vector/insert ratio of 3:1 is reqiured to generate the most recombinants because its not simply a ratio directly to be taken in the ligation it totally depends upon the size of both the vector and insert. Vector is alwys bigger in size that the insert and to produce a competitive number of DNA ends after digestion it is reccomende to use 3:1 ratio of both the componets.
