The lettered plates above resulted from a transformation experiment just like the one you performed in lab. You plated the same amount of bacterial suspension onto each plate. Use the letters of the plates to answer the following: Which plate(s) contain (s) ampicillin? Which plate(s) contain (s) arabinose? From which plate(s) would you collect bacteria if you were interested in getting clones of the gene for GFP?

Solution

Answer for question 19: The plate lettered B and C contains ampicillin, because in this experiment there will be 4 types of bacteria. So on the plate B the bacteria is with a fluid called lauria broth that is basically food for the bacteria along with ampicillin, as ampicillin is an antibiotic so bacteria may not be able to grow and that\'s why we cant see anything clearly, while on the plate C the bacteria is along with three component they are lauria broth, ampicillin and plasmid. So in such case the bacteria that picks up the plasmid will be able to grow and this is the reason that we can see some spots on plate C.

Answer for question 20: The plate that contains arabinose sugar is lettered as D. The bacteria present on plate D contains lauria broth, ampicillin and arabinose sugar. Because of their presence the bacteria were able to transform and due to the presence of arabinose sugar they were able to produce such glowing protein as arabinose sugar acts as a trigger.

Answer for question 21: I would collect bacteria from the plate lettered D because they have arabinose sugar which helps the bacteria to show up so nicely and to glow. And even due to the presence of the arabinose sugar the bacteria will become able to get the clones of the gene for GFP that is glowing fluorescent protein.
