The bands shown in the YFP, P1+P2, P3+P4 depends on the number of basepair in the DNA. YFP has low basepair compared to P1+P2 and P3+P4. The tritemplate PCR band is formed by fusing of 3 genes, YFP, P1+P2, P3+P4. This tri template PCR is used when there is no specific ligation site for an endonuclease. Three primers for this specific genes are used to elongate and fuse the 3 genes. To generate such a TT PCR band in elctrophoresis , 25 to 30 cycles is enough . Because the 3 fusion product will be around 4000 bp and to generate little more 25 cycles for 1 min is enough.
