Solution
After transferring the sample on to the slide, the smear is formed and this smear is to be fixed to allow the observation without disturbance (organism cannot move). If use the over-heat to fix the cell, the cells take the heat and it denature the proteins of the cell membrane. It may cause loss of certain morphological features, which cannot be observed.
It is important to make a thin smear because if we make the smear thick, the cells appear clumsy.
