Solution

1) Ethanol is antimicrobial which is used to sterilize the spreader used for spread plate technique. The spreader should be aseptic before it is used to take the inoculum because only the desired type of strain is allowed to grow.

2) In streak plate technique, a loop is used to take the inoculum and streaked in the plate to give some patterns. At the end of streaking, individual cells from the loop will be removed to form individual colonies.
