The bacteria on a hanging drop slide are not stained. Unstained bacteria have very little contrast. What single adjustment would you make to your microscope that would make the cells easier to see?
Solution
While focussing the microscope, it is necessary to reduce the light when observing unstained bacteria. This will allow for better visibility of the microorganism.
Using Differential Interference microscope and Bright field microscope is useful for unstained specimens.
