stop and think
You will incubate with an antibody against one of two proteins or both: anti-rabbit zonula occludens-1 (ZO-1), a protein that localizes to the cytoplasmic membrane surface of intercellular tight junctions of epithelial cells. ZO-1 is involved not only in junction complexes, but also in signal transduction at cell-cell junctions; anti-mouse smooth muscle actin (SMA), Actins are highly conserved proteins that are involved in cell motility, structure and integrity. Alpha actions are a major constituent of the contractile apparatus in smooth muscle cells. Stop and Think: What species are the antibodies raised in? What are the species-specific binding characteristics for each antibody? What is the molecular explanation for species-specific binding? What are the important considerations with regard to secondary antibody choice when staining with one or both of these antibodies?Solution
antibodies can be raised in any other species other species like anti rabbit can be raised in other species human horse donkey etc and so applies to anti mouse .....
anti rabbit binds to rabbit immunoglobulins (protiens)
where as anti mouse binds to mouse immunoglobulins(protiens)
secondary antibody must bind to the primary antibody and must contain the label conjugate for detection (colorimetric, flourescent, radioactivity ) or other labels so that we can find where primrary antibody binds
Hope it helps feel free to discuss more
