Compare the appearance of microorganisms you observed using phase-contrast microscopy with that of those you observed using bright field microscopy?
Solution
phase contrast microscope makes the specimen to look brighter or darker . This change in phase of light is dependent on the optics which is used, thus their is an increase or change in the color contrast with the mounting medium. Whereas in bright field microscope the color of the specimen is not changed. It appears in same color as when mounting. The specimens are generally stained before mounting on the bright field microscope if they are not originally colored specimens.
