Describe what would be seen under a brightfield vs darkfield
Describe what would be seen under a brightfield vs darkfield microscope viewing fibers as the specimen. What are some of the characteristics and descriptions of the fibers.
Solution
Bright field microscopy is the simplest way of techniques used for illumination of samples under light microscope whereas in darkfield microscopy complex multiple condensor system having reflecting surfaces at specific geometries is present.
Under darkfield microscopy fibres appears as brilliant, light images against a dark background whereas in Bright field microscopy the object is lit from below the stage, resulting in a larger, contrasted image.
In brightfild microscopy sample contrast comes from absorbance of light in the sample whereas in case of darkfiled sample contrast comes from light scattered by the sample.
Different fibres posssess different characteristics however some of the characteristics includes
1) Absorbency : which includes ability to regain moisture.
2) elasticity : all fibers do not have the same breaking point so its elasticity differs and it is the ability to recover from the strtch.
3) heat sensitivity : wool, silk and all proteins if boiled then it results into damge to its structure.
4) wrinkle recovery : in case of relaxed fibres wrinkles appears in less a manner.
5) chemical resistance : thye fibre should withstand at different pH level.
