If you are provided with a CHO cell line expressing an adrenergic receptor, how could you experimentally demonstrate that this receptor coupled to increases in cAMP via a G protein?

Solution

Ans. ) CHO cells lines are basically epithelial cell line originated from the ovary of the Chinese hamster. It is used in  medical research for instance in the commercial production of several therapeutic proteins.

These CHO cell lines are able to mediate a significant increase in intracellular cAMP level. In these cells, histamine H2 receptors enhance the basal cAMP level and also improve forskolin-induced cAMP production. The signaling of endogenously and transiently expressed G- protein coupled receptors (GPCRs) reduced proportionally to human H2 receptor expression levels. The G protein transfection specifically receded this attenuation and suggests seizing of the G protein from a common pool. The concentration-response curves carried out with the specific H2 agonist amthamine also showed an increase in cAMP level.
