Please help with the follwing True or False?
a) ATP is a substrate of PFK?
b) ATP is a competitive inhibitor of PFK?
c) ATP is an allosteric inhibitor of PFK?
d) An increase in PFK activity leads to an increase in cellular ATP production?
Solution
True: ATP is a substrate of PFK. There are 2 binding sites for the ATP in the enzyme such as binding site for the substrate and one for the inhibitory site. The substrate binding site of PFK links ATP well in T as well as in R state. False: ATP is a non competitive inhibitor of PFK. Accumulation of ATP results in it binding to a site which is other than the active site on PFK True: ATP is an allosteric inhibitor of PFK as in a high ratio of ATP when compared to ADP would inhibit PFK False: an increase in PFK activity would only reduce the cellular ATP and they are inversely proportional as the ATP gets used up.
