1.What was the concentration (cell/ml) of your final dilution in your growth curve experiment? How many colonies do you expect to see on the LB plate? Show your calculation(s)! My 37* C E.coli culture was 0.678 and predicted OD(optical density) was1.356 [on plate A spread 10 ul of the last dilution and 90 ul of LB. On plate B spread 100 ul of the last solution, and on plate C spread 100 ul of the dilution before the last.
Solution
You\'ve got 356 cells/mL in the last dilution.
10 uL is 1/100(th) of 1 mL, so you would expect 1/100(th) the number of cells.
(1/100) * 356 = 3.56, or approximately 4 colonies.
100 uL is 1/10(th) of 1 mL, so you would expect 1/10(th) the number of cells.
(1/10) * 356 = 35.6, or approximately 38 colonies.
