Mature mRNA from eukaryotic cells is often purified from other components in the cell with the use of columns containing oligo (dT) cellulose. These columns contain short segments of ssDNA of deoxythymidine bound to a cellulose matrix. Explain the rational for use of these columns to purify mature mRNA from a mixture of components.

Solution

The main rationale beg=hind the extraction of mRNA from a mixture of components using oligi dT columns, is that the mRNA that will be obtained at the last of the reaction has to be ultra pure. It should not be contaminated with any other components like DNA or Proteins.

To increase the abundance of pure mRNA , the method is used wich ensures separation of around 95% pure mRNA.

The presence of poly- A tail at the 3\' end of the mRNA makes this method , the most useful one. this is around 50 300 nucleotide long poly A tail. To this Poly A tail, the oligo dT column will bind, leaving behind every molecule which donot have polyadenylated tail.

So after elution pure mRNA will be obtained and other contaminants will be left behind.
