A. You are given 50 µL of a highly concentrated DNA solution. You need to prepare 1.0 mL of a 1:500 dilutions. Describe how you would prepare this solution

B. You diluted a DNA solution 1:100 and measured the OD at 260 nm. The spectrophotometer gave you a reading of 0.35. Describe how would you determine the DNA concentration of your original solution? Remember, the DNA = 50x10-3 mg/mL.

we will apply the molarity formula

M=(Given mass * 1000)/ (molecular mass * volume of solutions(ml))

A) according to this formula,

M=1.5 M

Volume of solutions =500 ml

Molecular mass = as given..

We can calculate the given mass required to dissolve in this amount of water to obtain the 1.5M... To that if we 100 gm of KCl we will get the desired solution..!!

B) similarly we can also find the weight of the NaCl required to dissolve in 0.75L to obtain 50mM...!! To that add 75 gm of SDS..!!

C) with the above given formula, we can put the value in the formulae we can calculate the given mass required to dissolve in this amount of water... Add 1gm of agarose in it..
