Explain how each of the following would affect the final result of a simple stain. Using a large amount of bacteria Using India Ink instead of Methylene Blue. Using Gram violet stain rather than Methylene Blue.

Solution

a. using a large amount of bacteria would cause a crowdy picture in stain, therefore if it is cocci it would be quite difficult to identify the arnngment of cocci. A cluster may indicate group while it is not so.

b. india ink is negative stain which do not stain capsule.so bacteria will not take colour but onle the caspule and there would be hollow around capsulated bacteria where as non-capsulated bacteria presence would be neglected.

c. Methylene Blue, Loefflers is recognized as a simple stain used for determining bacterial morphology and negatively charged molecules in the cell causes the staining phenomenon. Gram crystal violet used in gram stain where it forms a complex with iodine to give stain. So as a simple stain crystal violet has no use in differentiation of bacteria how ever it would give a blue purple colour to bacteria.
