In molecular cloning, how do we ensure the bacteria cells we select have taken recombinant plasmid?

Solution

There are various methods like we can use bluewhite screening, tag bacteria with GFP ...or simply we can use antibiotic selection ...

Suppose the plasmid contaning protien of intrest also has an antibiotic resistance say ampicillin resistance gene, we then grow the transformed culture into an ampicilin contaning medium... what we will find is that only those cells will grow which contain plasmid because antibiotic resistance gene is on plasmid. and other cells without plasmid will die without resistance gene. In this way we would get only those cells which got sucessfully transformed with plasmid.
