(b) (5 pts.) You then do an experiment to measure reaction velocity as a function of substrate concentration and obtain the results shown below 15 210 0.5 1.5 2.5 Substrate Concentration (mM) From this graph, estimate Km and Vmax, with appropriate units

Solution

b. Vmax is the Maximum velosity of an enzyme when it is active sites are fully saturated or fully occupied. When we take the half of its maximum velocity, then it is called Vmax /2. The substrate concentration at Vmax /2 is called Michale Mentons Constant i.e. Km. Based on the graph shown abobe Vmax is nearly 15uM/min & Km is nearly 0.3 mM.

c. K cat = V max / Total Enzyme

= 15uM/min/3ug/ml

= 15 x 10-6 M/min / 3 x 10-6 g/ml

= 15 M/min / 3g/ml

= 5/min

 (b) (5 pts.) You then do an experiment to measure reaction velocity as a function of substrate concentration and obtain the results shown below 15 210 0.5 1.5
