mammalian vectors have

kozak sequences and sv40 origin of replication..... as mammals are eukaryotic organisms and kozak sequence is only found in eukaryotic transcripts ..... where as prokaryotic have TATA box consensus promoters. Similarly simian virus 40 (sv40) promoter wors only in mammals... as derived from simian virus which infects humans (mammals)

where as in ecoli

f1 origin of replication from phage infecting ecoli

similarly COLE1 is form bacterial plasmids andstands for colicin E1 gene .....

gfp stands for green flourescent protien ... it glows(shows flourescence) when exposed to light in the blue to ultraviolet rang and is attached to protien sequence as shown above, so where ever there is protien in cell that part glows up and that glowing region shows us the localisation of protien within cellular organeles ....where as myc tag doesnt glow so cannot show localisation ..... so vector with gfp can be used for localisation/location studies .....hope it helps
