Solution
According to the Microbiologists, the word inoculate means simply to introduce or insert one substance into another. So if bacteria is inoculaed into different media like broth, slants, plates, deeps etc, it is done purposefully to identify subsequent reactions of the introduction of particular bacteria in a soecific media.
we use Luria Broth which is a nutrient-rich media commonly used to culture bacteria in lab. The addition of agar to LB results in the formation of a gel that bacteria can grow on, as they are unable to digest the agar but can gather nutrient from the LB within. The addition of an antibiotic to this gel allows for the selection of only those bacteria with resistance to that antibiotic- usually conferred by a palsmid carrying the antibioytic resistance gene. The following protocol will allow one to make ones own LB/agar plates with youyr own antibiotic of interest as compare to LB slant.
