The N-terminal domain of p53 used to interact with basic transcription factors and transcription coactivators which are having acetyl transferases activity. transcription coactivators p300 and PCAF acetylate p53 C-terminal lysine residues such as K382. p53-binding protein SIRT1 specifically deacetylate p53 K382. p53 is acetylated at K382 and also at 320. A number of Lys residues at the C-terminus of p53 are the targets of both acetylation and ubiquitination.
The non-specific DNA binding to p 53 is due to three lysine residues (K373, K381, and K382) which are having positive charges and they confer the electrostatic binding, when these lysines are eliminated when their amino groups are acetylated
