1) True: A bacterial capsule is the outside layer of the cell which is also called as the cell envelope of the bacteria. It can be found in both gram-positive and gram-negative bacteria. It is usually made up of polysaccharides, also consists peptidoglycan and muramic acid. The capsule takes a lighter stain than the bacteria and its background during any microscopic observation. As it is tightly packed it is difficult to stain the capsule.
2) False: Bacillus species and E coli are gram positive bacteria and the gram positive bacteria always retains the primary stain i.e. crystal violet and will not absorb the secondary stain i.e. safranine and hence gives a violet or purple color stain when viewed under the microscope. and if the bacteria is a gram negative bacteria then it retains the secondary stain and loses the primary stain and appears as red when viewed under the microscope.
