A. COMPLEMENTATION vs STRAIN

Complementation occurs whe two strains of an organism with different homozygous recessive mutations that produce the same mutant phenotype produce offsprings with the wild type phenotype when mated or crossed. Complementation occurs only when the mutations are in different genes. In this case, each strains genome supplies the wild type allele to complement the mutated allele of the other strains genome. Since the mutations are recessive the offsprings will display the wild type phenotype. A complementation test also known as the cis-trans test is sometimes used to check whether the mutations in strains are in different genes. Complementation will not occur if the mutationa are in the same genes. Complementation test was founded by American Geneticist EDWARD B. LEWIS.

YPD- is known as Yeast Extract Peptone Dextrose is a complete medium for yeast growth.It contains yeast extract, peptone, double distilled water and dextrose or glucose. It can be used as a solid medium by using Agar. The yeast extract will typically contain all the amino acids for the growth. It cannot be use as a selective medium for the test of auxotrophs, rather its used as a growth medium for the yeast cultures.

SD media is the synthetic defined media, which includes the additional amino acids which may be required for the growth of an auxotrophic strain.
