In order to prepare TE buffer most people start with a 1 M solution of Tris which has been adjusted to a pH of 8.0 with a small amount of hydrochloric acid (HCl). The formula weight of Tris is 121g/mole. How much Tris powder would be needed to prepare 50 ml of a 1M solution? Please show your work.

Solution

Amount of Tris powder required = (gram MW of chemical) x (volume needed in Liters) x (final concentration in Molarity)

Gram molecular weight of Tris = 121g/mole

volume in liters = 50 / 1000 = 0.05

Final concentration = 1M

Amount = (121) (0.05)(1) = 6.05 grams.

Therefore, to prepare 50ml of 1 Tris solution, 6.05 grams of Tris powderis required.
