You have just finished an exhausting genetic screen. You now want to fully characterize one new mutation you are calling mtp. You need to determine if the mtp mutation is a nullomorph, hypomorph, or hypermorph. Describe multiple types of experiments you would perform to fully characterize this new mutant (number them for clarity) AND describe your interpretation of possible results for each experiment using the following type of sentence structure/ logic: If I do __ and see _____, then I will conclude_______. In contrast, if I see____, then I will conclude ___________)
Solution
If I do deletion of “mtp gene” and see excess production of gene products (protein), then I will conclude then the mutation is hypermorph. In contrast, if I see reduced production of gene products (protein), then I will conclude that the mutation is hypomorph.
If no result occurs, after deletion the gene mutation, that means that it is a nullomorph.
Qs. Describe multiple types of experiments you would perform to fully characterize this new mutant
As
1. Molecular genetic tests: In these, single genes or short lengths of DNA are studied. Deletion or addition mutations are performed for analysis of results.
2. Chromosomal genetic tests: In these, whole chromosome is taken into consideration. Defects which occur due to whole chromosome addition are studied in this. Ploidy related effects also come in this type of genetic screening method.
3. Biochemical genetic tests: These tests rely on activity of a single protein/enzyme in solution. Please note that one gene may control the effect of more than one enzymes. So, biochemical tests are very specific, and give the exact abnormality related with the disease. A biochemical test can be tracked back to genes and alteration in DNA which had led to the particular diseased condition.
** In the above analysis, I have written the answer on the basis of chromosomal tests.
