For each of the following mutations, speculate on how the mutation would affect the ability of nucleosomes to assemble. Be specific on which step of the assembly process would be disrupted. mutation that prevents the histone H2A histone fold domain from forming. mutation that deletes the alpha helix in the N-terminal tail of histone H3. Grading: For each: 1 pt for correctly and specifically identifying the assembly step that is disrupted 1 pt for explaining why that mutation disrupts that assembly step.
Solution
Mutation in H2A prevents the dimerization process of H2A to H2B. That will cause to stop the further step of assembly i.e. H2A-H2B dimer to H3-H4 trimer assembly.
Mutation that deletes the alpha helix H3 will stop the first step of nucleosome assembly. First step is the formation of H3-H4 tetramer. Deletion of alpha helix will lead to stop the process and then H3-H4 tetramer would not deposited over the DNA.
