I am doing a research, I need an scientific assay for the Urease enzyme activity.

Solution

Urease is an enzyme that convert Urea into Carbon dioxide and Ammonia.

The enzyme activity can be determined by measuring the amount of Amnonia produced in the reaction. Since Ammonia forms a brown colour complex in the presence of Neissler\'s reagent this assay can be done colorimetrically.

1. 3% Urea

2. 66N Sulfuric acid

3. 1M Sodium tungstate

4. 2M Phosphate buffer (pH7)

5. Neissler\'s reagent

6. Ammonium sulfate
